The archive was organized during two periods of activity: by Burroughs in 1965, and then in 1972 by Burroughs, Brion Gysin, and Barry Miles, who divided the manuscripts and typescripts of Burroughs's works and notes into 169 collections of papers which they placed in consecutively numbered folders and portfolios of various sizes, which they called "Folios". Burroughs inscribed headings on the folders and sometimes also affixed collages to their front covers. In addition to manuscripts and typescripts, these folios include correspondence, clippings from magazines, photographs, exercise books, and cut-ups. Truman Capote (1924-1984) was a celebrated American author whose major works include
Other Voices, Other Rooms;
Breakfast at Tiffany's; and
In Cold Blood. John O'Shea (John Matthew O'Shea, 1929-2014) was a married Long Island banker who became Truman Capote's lover and business manager in the early summer of 1973. The Truman Capote papers regarding "Houston Diary," 1973-1974, consist of materials pertaining to Capote's writing interest in the mass murders committed in Houston, Texas by Dean Corll, discovered in August 1973, and his intended coverage of the 1974 trial of Elmer Wayne Henley, one of Corll's accomplices. Capote's trial coverage was to be syndicated by the Washington Post under the title "Houston Diary," but the project did not come to fruition. The collection, created in part and apparently maintained by John O'Shea, contains letters received by Truman Capote from members of the press; two holograph manuscripts by Truman Capote relating to the
Washington Post project; O'Shea's business papers concerning publishing negotiations, chiefly with the Post; a scrapbook of newspaper and magazine clippings; loose clippings; and photographs. Correspondents include
Washington Post editor Ben Bradlee and
Village Voice journalist Arthur Bell.

The Richard Barr-Clinton Wilder Papers contain a great deal of documentation on the working aspects of the collaboration between Barr, Wilder and Edward Albee whose various producing units helped to establish a number of important American playwrights in the 1960s. In addition they document the work that Barr did with Charles Woodward, Jr. after Wilder left the Playwrights Unit as well as the work done by Barr and Wilder before they formed their partnership. The Administrative Files and Production series contain correspondence; production files, such as contracts, agreements, cast and crew lists, inventories, schedules, costume plots, and lighting plots; some scripts; programs; legal papers; business papers and clippings. The bulk of the documents, however, are financial records including royalty statements, box office statements, ledgers, bank statements, invoices and receipts. The more personal papers in the collection including writings, photographs and ephemera document Richard Barr's life and work. There are no similar items documenting the life of Clinton Wilder. The scrapbooks document the work of the Playwrights Unit, mostly through clippings; they appear to have been put together by Barr.

The papers of Lucille Lortel relate the details of her life and career from teen years to her death in 1999, and include correspondence, production files, scripts, programs, production photographs, personal and family photographs, organization files, clippings, memorabilia, and scrapbooks. Lucille Lortel's life spanned the twentieth century, so in addition to providing details of her family and personal life her papers encompass many aspects of the theatrical history of her era. Lortel is credited with fostering the Off-Broadway movement and providing a forum for avant-garde and experimental work at her Theatre de Lys. Lortel's productions at the White Barn and the ANTA Matinee Series at the Theatre de Lys brought works by Jean Genet, Sean O'Casey, Athol Fugard, and others to a wider audience. Many of these productions are represented in the collection by correspondence, programs, photographs and clippings. Over the years Lortel also worked closely with several non-profit theaters as a donor and mentor. Her affiliations with Circle in the Square, Circle Repertory Company, Goodspeed Opera House, Yale Repertory Theatre, and other companies are documented in the organization files.
